Mutational spectrum of DNA mismatch repair genes MLH1, PMS2 and PMS2 in patients with sporadic colorectal cancer in Thailand.
A large proportion of hereditary nonpolyposis colorectal cancers (HNPCC) are caused by mutations in DNA mismatch repair (MMR) genes. We sought to determine the prevalence of MMR gene mutations in Thai patients with sporadic colorectal cancer. The patients were selected at the Queen Sirikit National Institute of Child Health (QSNICH), Bangkok, Thailand. We used a PCR-SSCP approach for the detection of MMR gene mutations in DNA extracted from peripheral blood leukocytes from patients with colorectal cancer. Mutations were identified in 16 of 61 (26.2%) patients; PMS2 and MLH1 mutations were found in eight and eight patients, respectively. We have demonstrated a relatively high frequency of MMR gene mutations in Thai patients with colorectal cancer.
